Description
| - The project goal is a comparative study of bioenergetics pathways, signaling mechanisms and membrane functions of thawed sperm during motility observation using chondrostean and teleostean fishes. We will demonstrate that according to process of cryopreservation and thawing of sperm, membrane permeability and dyneins/microtubules reactivity flagellum would become changed. On the base of these studies we will try to find possible species specific proteins responsible for flagellar regulatory mechanisms at chondrostean and teleostean fishes and find correlation between thawed motile sperm and protein structure of the flagellum. Firstly we will try to understand and to explain why duration of motility is affected after cryopreservation by evaluation of ATP, ADP and PCr level and also cryopreserved demembranated spermatozoa with studdying cryobiology machinery of microtubules with midepiece in both model species. - Cílem projektu je komparativní studie bioenergetických drah, signálních mechanismů a funkce membrány v průběhu motility rozmrazených spermií u chrupavčitých a kostnatých ryb. Budeme se snažit ukázat, že na základě zmrazení a rozmrazení spermií dojde ke změnám permeability membrány, včetně dyneinové-mikrotubulové reaktivity bičíku. Pokusíme se najít druhově specifický protein regulující pohyb bičíku spermií u chrupavčitých a kostnatých ryb a rovněž nalézt korelaci mezi motilitou rozmrazených spermií a proteinovou strukturou bičíku u těchto druhů. Poprvé se budeme snažit u obou modelových druhů vysvětlit proč doba motility spermií je ovlivněna zmrazováním a to vyhodnocením úrovně ATP, ADP a PCr a rovněž se pokusíme zmrazit demembranované spermie pro studium kryobility mechanismu funkčnosti mikrotubulů, včetně střední části spermie.

| - Sperm physico-chemical properties, protein profiles, cryoresistance and fertilizing ability were studied in sturgeons and bony fish species. For five years was published 31 articles in journals with IF from 0.7 to 3.3 and a chapter in the book. (en)
- Fyzikálně-chemické vlastnosti spermií, jejich proteinové profily, cryoresistance a oplozovací schopnost byla studována u jeseterů a kostnatých rybích druhů. Za pět let bylo publikováno 31 článků v časopisech s IF od 0,7 do 3,3 a jedna kapitola v knize. (cs)
