Description
| - The cells in multicellular organisms are spatially organized through cell-cell and cell-extracellular matrix interactions. Disruption of adhesive bonds results in enhanced cell motility and helps cells to become invasive - to degrade the basal membrane,escape from their home tissues, invade surrounding tissues and eventually to colonize them. Cell invasiveness plays an important role in the normal embryonic development and also in metastasis - the most dangerous stage of tumours, which is the actual cause of death in approximately 90% of cancer patients. In both cases the cell invasiveness appears to be regulated by similar mechanisms, which are presently known only partially. In order to contribute to the knowledge of cell invasiveness regulation wewill study in parallel selected molecules and molecular mechanisms that regulate cell migration and invasiveness in early vertebrate embryos and in cells of non-invasive and metastasizing experimental tumors. - Buňky mnohobuněčných organizmů jsou poutány vazbami jedna k druhé nebo k extracelulární matrix, která vyplňuje mezibuněčné prostory. Uvolnění těchto vazeb má za následek zvýšenou pohyblivost buňky a případně i invazivitu - schopnost opustit původní strukturu tkáně, proniknout přirozenými bariérami a případně kolonizovat nové prostory. Buněčná invazivita se významně podílí na normálním vývoji embrya, ale také na nejnebezpečnějším stadiu zhoubného nádoru - metastáze. V obou zmíněných případech je invazivita zřejmě regulována stejnými mechanizmy, jejichž znalost je však zatím pouze částečná. Studiem vybraných molekul a molekulárních mechanizmů regulujících buněčnou invazivitu v časném embryu obratlovce a v buňkách experimentálních nádorů budou určeny ty molekuly, které se na invazivitě buněk významně podílejí. Získané poznatky budou ověřeny na experimentálních nádorech in vivo. Rovněž budou analyzovány lidské nádory, primární i metastázy, se snahou určit, které ze zjištěných molekul se mohou podílet na v

| - Centrum buněčné invazivity spojilo výzkumné úsilí molekulárních biologů, lékařů a organických chemiků z ÚMG a ÚOCHB AV ČR a Karlovy Univerzity. Invazivita nádorových buněk byla pochopena do té míry, že byla cíleným zásahem do její regulace vyvolána schopnost metastázy. Byly též hlouběji poznány různé typy buněčného pohybu a jejich regulace a připraven originální soubor syntetických látek. (cs)
- The Center of Cell Invasion brought together biologists, physicians and chemists from IMG, IOCB ASCR and Charles University. Understanding the molecular mechanisms of metastasis enabled manipulation with metastatic phenotype of experimental tumor cells.
