  • Protoonkogen c-myb i z něho odvozený onkogen v-mybAMV kódují transkripční regulátory schopné ovlivnit komitment krevních progenitorových buněk. Na rozdíl od proteinu c-Myb, který působí při tvorbě a diferenciaci různých typů progenitorů, onkoprotein v-Myb komituje myeloidní progenitor do linie monocytu a způsobuje jeho transformaci ve stadiu monoblastu. V tomto článku poprvé dokazujeme, že bílkoviny Myb určují buněčný osud i v buňkách neurální lišty, které směrují do linie melanocytu. V buňkách neurální lišty bílkoviny Myb aktivují expresi receptoru c-kit, čímž zřejmě aktivují signál tohoto receptoru. Jak bylo dříve ukázáno, je exprese c-kit v erythroidních progenitorech rovněž závislá na c-Myb. Zdá se tedy, že signální dráha c-myb - c-kit představuje regulační signál společný pro vývoj krevních i neurálních progenitorů. V této práci zároveň představujeme nový experimentální model pro studium melanocytogeneze a transformaci melanocytů. (cs)
  • The c-myb proto-oncogene and its oncogenic derivative v-mybAMV encode transcriptional regulators engaged in the commitment of hematopoietic cells. While the c-Myb protein is important for the formation and differentiation of various progenitors, the v-MybAMV oncoprotein induces in chicks a progression and transformation of the single (monoblastic) cell lineage. In this article the evidence is presented that c-Myb and v-MybAMV proteins have cell fate-directing abilities also in avian neural crest (NC), where both proteins determine the melanocytogenesis. Both Myb proteins activate in NC cells expression of the c-kit gene and the stem cell factor-c-Kit signaling – one of the essential pathways in melanocyte development. These observations bring evidence that the c-myb-c-kit pathway represents a common regulatory scheme for both hematopoietic and neural progenitors and establish a novel experimental model for studies of melanocytogenesis and melanocyte transformation.
