| Description
| - One of the main directions of human genome research in the post-genomic eta is the study of its higher-order structure that became the key to understanding of the most important cellular processes such as cell differentiation or transformation. Using high-resolution cytometry developed in our laboratory, nuclear and territorial topography of genetic elements will be systematically investigated in normal and malignant human cell types. The topography and its changel will be correlated with local acetylation of histones, methylation of DNA and expression of gene clusters. In addition, changes of the structure of chromosome terriories or whole human genome in the cell nucleus during differentiation and cancerogenesis will be determined and an attempt will be made to find the basic psrinciples of the epigenetic regulation of gene expression. The practical goal of the project is the use of experimental results and technology developed in our laboratory in diagnostics ant therapy of cancer. (en)
- Po rozluštění genetického kódu je jedním z hlavních směrů výzkum lidského genomu studium jeho struktury vyššího řádu, které se stalo klíčem k pochopení nejdůležitějších buněčných procesů jako je diferenciace či transformace buňky. V tomto projektu bude systematicky zkoumána jaderná a teritoriální ropografie různých genetických elementů ve zdravých i nádorových buňkách s využitím cytometrie s vysokým rozlišením, vyvinuté v naší laboratoři. Topografie a její změny budou korelovány s lokální acetylací histonů, metylací DNA a expresí genových klastrů. Kromě toho budou studovány změny struktury chromosomových teritorií a buněčného jádra během diferenciace a kancerogeneze. Na základě získaných poznatků se budeme snažit o nalezení základních principů epigenetické regulace exprese genů. Praktickým cílem projektu bude využití získaných poznatků v diagnostice a při léčbě nádorových onemocnění.

| - In the project, we have investigated the arrangement of the human genome. Correlation between the arrangement of the genome and its function, particularly transcription, has been found. A model of the genome arrangement has been proposed. (en)
- V rámci projektu jsme se zabývali strukturou lidského genomu. Byla získána řada významných výsledků svědčících o tom, že funkce geonomu, zejména jeho transkripce, koreluje s jeho uspořádáním v jádře buňky a byl navržen model tohoto uspořádání genomu. (cs)
