Many different signalling pathways that monitor the dynamic state of the cell converge on p53. The ability of p53 to process these signals and respond by triggering cell cycle arrest and/or cell death by apoptosis is crucial for prevention of tumour development. P53 is mutated in more than 50% of cancer cases. One of the future strategies for treatment of cancer is based on drug-mediated reactivation of mutant p53 protein. Our project is focused towards biological, biochemical and biophysical studies of p53 and its homologues and their interactions with DNA to understand better multiple functions of these proteins in a cell. The role of post-translational modifications of p53 protein and the response of cells carrying different mutant forms of p53 to DNA damaging agents as well as to other types of cellular stress will be investigated. Mnohé signální dráhy, které monitorují dynamický stav buňky, směřují k proteinu p53. Schopnost p53 zpracovat tyto signály a odpovědět na ně zástavou buněčného cyklu a/nebo programovanou buněčnou smrtí je nezbytná pro prevenci vývoje nádorových onemocnění. Vzhledem k tomu, že protein p53 je mutován u více než 50% lidských zhoubných nádorů, je jedna ze strategií léčby nádorových onemocnění založena na reaktivaci mutovaných forem tohoto proteinu. Předkládaný projekt je zaměřen na biologické, biochemické a biofyzikální studie proteinu p53 a jeho homologů a jejich interakce s DNA s cílem pochopit mnohačetné funkce těchto proteinů v buňce. V průběhu řešení projektu bude studována úloha posttranslačních modifikací proteinu p53 a odpověď buněk exprimujících různé mutované formy tohoto proteinu na vyvolané poruchy DNA a buněčný stress. Studie bude zahrnovat rovněž biochemickou a biofyzikální analýzu interakcí proteinu p53 a jeho homologů se superhelikální DNA. Získané výsledky tohoto projektu
