Description
  • Cascade reactions represent the current trend in biocatalysis as means to improve its economic and ecological viability (due to no need for intermediate isolation, better product purity, racemization of substrates or regeneration of cofactor in situ). The project aim is to examine the possibilities of multistep catalysis in nitrile biotransformations leading to high-value carboxylic acids (nicotinic, isonicotinic, mandelic, lysergic etc.) and hydroxamic acids (chelators). The action of nitrilases and amidases will be integrated to produce high-purity carboxylic acids from nitriles (the role of amidase being hydrolysis of the nitrilase by-products, amides). Nitrile hydratases (or mild chemical catalysts) and amidases from different strains will be combined to give carboxylic acids from nitriles, if enzymes produced by single strain are not compatible in terms of substrate specificity.   • Kaskádové reakce katalyzované chemicky a enzymy (nitrilhydratasami, amidasami, nitrilasami, lipasami) byly použity k přípravě farmaceutických produktů v laboratorním měřítku (cyklofelitolů - inhibitorů glukosidas, hydroxamátů - chelatačních činidel), intermediátů přírodních látek nebo semisyntetických antibiotik aj. Princip kaskádových reakcí byl využit také ke stanovení těchto enzymů. (cs)
