  • Real-time tomography is a novel quantitative method for measuring localized RNA-expression profiles within single cells. We show its usefulness by dissecting an X. laevis oocyte to slices along its animal–vegetal axis, extracting RNA and measuring levels of 18 selected mRNAs by real-time RT-PCR. This identified 2 classes of mRNA, one preferentially located towards the animal, the other towards the vegetal pole. mRNAs in each group show comparable intracellular gradients, suggesting they are produced by similar mechanisms. Polarization is substantial, though not extreme,with ca 5% of vegetal gene mRNA molecules at the animal pole and ca 50% in the far most vegetal section. Most animal pole mRNAs were found in the 2nd section from the animal pole and in the central section where the nucleus is located. mRNA expression profiles did not change after in vitro fertilization; we conclude that the post-fertilization cortical rotation has no detectable effect on intracellular mRNA gradients.

  • Real time tomografie je nová kvantitativní metoda pro měření lokalizovaných RNA-expresních profilů uvnitř jediné buňky. Demonstrujeme její použitelnost řezáním vajíček z X. laevis podél animálně-vegetativní osy, extrakcí RNA a měřením hladiny 18-ti vybraných mRNA pomocí real-time RT-PCR. To identifikovalo 2 skupiny mRNA, jednu přednostně lokalizovanou k animálnímu a druhou k vegetativnímu pólu. mRNA z obou skupin ukazují porovnatelné vnitrobuněčné gradienty, což může znamenat, že jsou produkovány podobným mechanismem. Polarizace je značná, avšak ne extrémní, přičemž okolo 5% vegetativních mRNA molekul je detekováno v animálních řezech a okolo 50% molekul v posledním vegetativním řezu. Většina animálních mRNA byla nalezena v druhých řezech od animálního pólu a v centrálních řezech, což je oblast výskytu jádra. mRNA expresní profily se nezměnily po in vitro oplození a z toho odvozujeme, že kortikální rotace, která se objevuje po oplození, nemá žádný vliv na vnitrobuněčné mRNA gradienty.
