Description
  • Syncytin-1 is a captive envelope glycoprotein encoded by one of human endogenous retroviruses W. It is expressed exclusively in the placental trophoblast where it participates in cell-to-cell fusion during differentiation of syncytiotrophoblast. In other tissues, however, syncytin-1 expression must be kept in check because inadvertent cell fusion might be dangerous for tissue organization and integrity. We describe here an inverse correlation between CpG methylation of syncytin-1 5' long terminal repeat and its expression. Our data on syncytin-1 methylation in the placenta, other primary cells and cell lines non-expressing syncytin-1 document that CpG methylation plays a principal role in the transcriptional suppression of syncytin-1 in non-placental tissues, and, in contrast, demethylation of the syncytin-1 promoter in trophoblast is a prerequisite for its expression and differentiation of multinucleated syncytiotrophoblast.

  • Syncytin-1 je obalový glykoprotein kódovaný jedním z lidských endogenních retrovirů W. Je exprimován výhradně v placentálním trofoblastu, kde se účastní splývání buněk při diferenciaci syncytiotrofoblastu. V jiných tkáních však musí být exprese syncytinu-1 přísně regulována, protože náhodné fúze buněk by mohly ohrozit organizaci a integritu tkání. Popisujeme zde opačný vztah metylace CpG 5' dlouhé koncové repetice syncytinu-1 k jeho expresi. Naše údaje o metylaci syncytinu-1 v placentě, jiných primárních buňkách a buněčných liniích neexprimujících syncytin-1 ukazují, že metylace CpG hraje zásadní roli v potlačení transkripce syncytinu-1 v neplacentálních tkáních a naopak, demetylace promotoru syncytinu-1 v trofoblastu je základním předpokladem pro jeho expresi a pro diferenciaci mnohajaderného syncytiotrofoblastu.
