Description
| - Using the model tick Ixodes ricinus, we have shown that host hemoglobin degradation in tick gut homogenates requires acidic conditions. A functional activity scan demonstrated the presence of five types of peptidases of the cysteine and aspartic classes in the gut of feeding females. We followed up with genetic screens of gut-derived cDNA to identify and clone genes encoding the cysteine peptidases cathepsins B, L and C, an asparaginyl endopeptidase, and the aspartic peptidase, cathepsin D. RT-PCR tissue expression profile displayed that all these proteases are highly expressed in the gut of feeding ticks and that the asparaginyl endopeptidase and cathepsins B and D messages are restricted to gut tissue and to those developmental stages feeding on blood. Overall, our results demonstrate the presence of an evolutionary conserved network of gut-associated cysteine and aspartic peptidases orthologous to those described in other parasites, including nematodes and flatworms.

- Použitím modelového klíštěte Ixodes ricinus jsme dokázali, že degradace hemoglobinu v extraktu střev sajících klíšťat vyžaduje kyselé prostředí. Profil hydrolytických aktivit pomocí specifických substrátů a inhibitorů demonstruje přítomnost pěti cysteinových a aspartových proteáz ve střevě sajících samic. Amplifikací a klonováním střevní cDNA se podařilo získat sekvence kódující cysteinové peptidázy cathepsiny B, L a C, asparaginylovou endopeptidázu a aspartovou peptidázu, cathepsin D. RT-PCR profil ukázal, že všechny tyto peptidázy jsou exprimovány ve střevě sajících samic a dále že mRNA kódující asparaginylovou endopeptidázu a cathepsiny B, D jsou exprimovány pouze ve střevě a ve vývojových stádiích klíštěte, které sají na hostiteli. Naše výsledky demonstrují přítomnost evolučně konzervovaného komplexu cysteinových a aspartových proteáz dříve popsaného u jiných parazitů, především nematod a krev sajících ploštěnek. (cs)
