Description
| - The main aim of the proposed project is to optimize the unconventional methods of hybridization via protoplast fusion techniques in economically important cultivated crops of the genus Brassica, Cucumis and Solanum. The methods of protoplast technology should help in developing the specific gene combinations in studied plant species, which are impossible to obtain through the conventional breeding approaches. Further this techniques offer a convenient way for overcoming the limitations on sexual hybridization in plant breeding. The goal of protoplast fusion is to control the transfer of an agriculturally important traits, such as disease resistance, abiotic stresses and also the improvement of the quality characteristics of seeds. The proposed investigations of this research are focussed on the development of the methods for symetrical and asymetrical protoplast fusion. - Cílem projektu je optimalizovat netradiční metody hybridizace pomocí fúze protoplastů u hospodářsky významných plodin. Metoda má přispět ke vzniku jedinečných kombinací genů různých rostlinných druhů, které není možné získat cestou klasického křížení a nabízí překlenutí sexuálních bariér ve šlechtění rostlin. Cílem fúzí protoplastů je přenos významných znaků rezistencí vůči patogenům, abiotickým stresům a kvalita semene. Cílem tohoto výzkumu je vyvinout metodiky pro symetrickou a asymetrickou fúzi protoplastů jak elektrickým polem, tak chemicky u zemědělských plodin a planých druhů rodu Brassica, Cucumis a Solanum. Vybrané genotypy jednotlivých plodin budou hodnoceny z hlediska výtěžnosti, životnosti a regenerace protoplastů. Průběh somatické hybridizace bude sledován cytologicky. Hybridnost produktů fúzí a případně získaných regenerantů bude ověřovány cytologicky a také metodami molekulární biologie. | - Řešení probíhalo v souladu s plánem projektu. V rámci rodů Brassica, Cucumis a Solanum byla fúzována řada kombinací, vedoucí u rodů Brassica a Solanum k regeneraci rostlin, u rodu Cucumis do stádia kalusů. Byla dosažena řada originálních výsledků. (cs)
