Description
  • Project intends to explore whether and to what extend cancer-related impaired interferon transduction pathways ensured by STAT 1 and SOCS 3 protein functions and interactions affect the response of malignant melanoma to IFN-alpha/gamma treatment The level of STAT 1/ SOCS 3 proteins, their mRNAs, and STAT 1 phosphorylated forms induced by IFNs in melanoma and normal cells will be examined using immunoblotting, MTT and morphological assays, and correlated with the interferon biological effects. It is expected to define newě markers which might predict responsiveness to interferon treatment in individual patients and to gain new data about IFN-induced STAT 1/SOCS 3 interactions in normal and malignant cancer cells.   • Projekt je zaměřen na analýzu patologických změn v přenosu interferonových signálů v nádorové buňce a jejich souvislosti s citlivostí nádoru na léčbu interferony. Na úrovni proteinů a mRNA studuje expresi transdukčního proteinu STAT 1, jeho interferony indukovaných fosfoforem a jeho interakce s inhibitorem SOCS 3 u maligního melanomu v korelaci s parametry v normální tkáni a s biologickými účinky IFN-alfa/gama. Metodickým základem je připrava imunosond proti SOCS 3 peptidu, Western a Northern blot, imunoprecipitace, MTT test a morfologická analýza buněčného poškození.
