Description
| - In obesity, the amount of excess fat and fat distribution show a large inter-individual variability. Differences in degree of adiposity account for approximately 25% of the variability in insulin sensitivity but substantially more of the variance is explained by differences in fat distribution. In the current project, we will perform genetic and correlation analyses of the role of ectopic fat accumulation and adipose tissue oxidative/inflammatory processes in the pathogenesis of insulin resistance in the BXH/HXB recombinant inbred strains derived from spontaneously hypertensive (SHR) and Brown Norway (BN) rats. Specifically, we will determine triglyceride levels in liver and muscle tissues, measure concentrations of selected adipokines and parameters of oxidative stress and sensitivity of adipose and muscle tissues to insulin action. - V předkládaném projektu provedeme genetické a korelační analýzy úlohy ektopické akumulace tuku a oxidačních/zánětlivých procesů v tukové tkáni v patogenezi inzulinové rezistence. Cílem projektu je odhalení odpovědných genetických determinant.

| - V průběhu řešení grantu byly u BXH/HXB rekombinantních inbredních (RI) kmenů odvozených od spontánně hypertenzních a Brown Norway potkanů odhaleny lokusy regulující adipozitu, ektopické ukládání triglyceridů ve svalech, sérové hladiny interleukinu 6 a leptinu. Dále jsme odhalili autokrinní účinky resistinu při inzulinové rezistenci a kauzální úlohu CRP v patogenezi metabolických poruch. (cs)
