  • We show that AC membrane translocation and pore-forming (hemolytic) activity of CyaA depends on a predicted transmembrane amphipathic α-helix, localized between residues 565 and 591 of the pore-forming domain and comprising pair of glutamate residues, Glu570 and Glu581. Moreover, we show for the first time that different amino acid substitutions of the glutamate residue at position 516 can increase the hemolytic activity of CyaA by two different molecular mechanisms

  • Ukázali jsme, že translokace AC domény a kanálotvorná (hemolytická) aktivita CyaA závisí na predikovaném transmebránovém amfipatickém α-helixu, který je lokalizován mezi aminokyselinovými zbytky 565 a 591 v hydrofóbní doméně a zahrnuje dva glutamátové zbytky, Glu570 a Glu581. Dále jsme poprvé ukázali, že různé aminokyselinové záměny glutamátového zbytku v pozici 516 mohou zvyšovat hemolytickou aktivitu CyaA dvěmi různými molekulárními mechanizmy (cs)
