  • Pro dosažení objemové rekonstrukce elasticky registrujeme konfokální obrazy z následných fyzických řezů. Využitím obrazů z USB světelného mikroskopu se přesnost registrace zvýší. Postup je následující: Nejdříve se registrují konfokální data vůči obrazům z USB mikroskopu a následně se registrují již přizpůsobená konfokální data vůči sobě. Výše popsaným způsobem dosahujeme dvojího cíle. Jednak kvantifikujeme míru deformací a tím chybu rekonstrukce a jednak na základě znalosti této chyby docilujeme přesnější objemové rekonstrukce. (cs)
  • We reconstruction results using a 17-day-old embryo of a laboratory Norway rat here. Ten confocal images of physical slices of the embryo were aligned with corresponding images of these slices captured by the USB light microscope by elastic registration. We measured areas of embryo slices from confocal data both before and after alignment and also from USB light microscope data. The measurements revealed partial deformations of cut physical slices (decreasing / increasing of areas measured from original confocal data with respect to USB light microscope data up to 92.7% / 105.3%), but an extensive one as well (decreasing the area of the embryo slice in confocal data to 55% only). However, even this dramatic deformation was successfully compensated by elastic alignment. Thus, our method for compensation of deformations using accessorial data from the USB light microscope turns up to be efficient and providing us with realistic visualization of biological structures under study.
