  • Na modelu retrovirového vektoru odvozeného z virů, které normálně vyvolávají nádory a leukemie u drůbeže, bylo zjištěno, že pro ochranu vektoru před inaktivací a methylací DNA je možno vložit dovnitř vektoru krátký úsek DNA pocházející z genů, jejichž aktivita je nutná v každé buňce a za každých okolností. Vektory vybavené takovými ochrannými prvky, tzv ostrovy CpG, zůstávají aktivní po dlouhou dobu a nepodléhají umlčování. Tato strategie by mohla vést k vývoji mnohem lepších vektorů (nejen retrovirových) pro přenos genů použitelných případně i pro genovou terapii. (cs)
  • Rous sarcoma virus and retroviral vectors derived from it suffer from progressive transcriptional silencing in mammalian cells. The insertion of the short core sequence of a CpG island into the long terminal repeat of such vector led to efficient protection of the integrated vector from silencing and gradual CpG methylation in rodent and human cells. Individual cell clones with IE-modified reporter vectors display high levels of reporter expression for a sustained period and without substantial variegation in the cell culture. The presence of Sp1 binding sites is important for the protective effect of IE, but at least some part of the entire antisilencing capacity is maintained in IE with mutated Sp1 sites. We suggest that this strategy of antisilencing protection by the CpG island core element may prove generally useful in retroviral vectors.
