  • Cnidaria is the earliest-branching metazoan phylum containing a well-developed lens-containing visual system located on specialized sensory structures, rhopalia. Each rhopalium in cubozoan jellyfish T. cystophora has a large and a small complex, camera-type eye with a cellular lens containing distinct families of crystallins. Here we characterized J2-crystallin and its gene in T. cystophora. Although nonfunctional in transfected mammalian lens cells, J2-crystallin promoter is activated by jellyfish paired domain transcription factor PaxB in cotransfection tests via binding to 3 paired domain sites. PaxB paired domain-binding sites were also identified in PaxB-regulated promoters of the J1A- and J1B-crystallin genes, which are not homologous to J2-crystallin gene. Crystallin recruitment of multifunctional proteins was thus driven by convergent changes involving Pax (and other transcription factors) in promoters of nonhomologous genes within and between species and within gene families.

  • Čtyřhranky jsou první odštěpující se živočišnou říší disponující velmi dobře vyvinutým vizuálním systémem obsahujícím čočku, nacházejícím se ve specializované smyslové struktuře nazývané rhopalium. Každé rhopalium medúzy čtyřhranky Tripedalia cystophora má velký a malý komplex, komorové oko s buněčnou čočkou, obsahující různé rodiny krystalinů. V této práci jsme charakterizovali J2-krystalin a jeho kódující gen v T. Cystophora. Přestože v transfekovaných savčích buňkách je J2-krystalin promotor nefunkční, je aktivován párovací doménou transkripčního faktoru PaxB medúzy v testech kotransfekce pomocí navázání k místům s třemi párovacími doménami. Místa navázání párovací domény PaxB byla identifikována také v PaxB regulujících promotorech genů pro J1A- a J1B-krystalin, které nejsou homologní ke genu pro J2-krystalin. (cs)
