  • Chemoterapeutika indukují jak zastavení dělení, tak apoptózu; ovšem některé rakovinné buňky se k nim stávají rezistentními. Potlačení imunitního systému chemoterapeutiky podporuje vývoj a množení různých malignit a také udržuje cytokinové prostředí favorizující dělení inhibováním apoptózy. Nový přístup je založen na synergistickém vlivu různých protirakovinných prostředků s různým způsobem působení. Nedávno byla pozornost zaměřena na redoxně-neaktivní analog vitaminu E, α-tokoferylsukcinát, na základě jeho protirakovinných vlivů. Vykazuje pleiotropní odpovědi v maligních buňkách vedoucí k zastavení buněčného cyklu, diferenciaci a apoptóze. Kromě jeho úlohy v zabíjení rakovinných buněk, α TOS ovlivňuje expresi genů na subapoptické úrovni. Zde shrnujeme současné poznatky o kooperativním protinádorovém vlivu α TOS, pokud je kombinován s imunologickými agens. Naznačuje to možnost využít novou strategii pro léčení rakoviny zahrnující zvýšený imunitní dozor. (cs)
  • Chemotherapeutic drugs induce both proliferation arrest and apoptosis; however, some cancer cells become resistant. The suppression of the immune system by chemotherapeutic agents promotes the development and propagation of various malignancies, and maintain a cytokine milieu that favors proliferation by inhibiting apoptosis. A novel approach is based on a synergistic effect of different anticancer agents with different modes of action. Recently, a redox-silent analogue of vitamin E, α tocopheryl succinate (α TOS), has come into focus due to its anticancer properties. It exerts pleiotropic responses in malignant cells leading to cell cycle arrest, differentiation, and apoptosis. Apart from its role in killing cancer cells, α TOS affects expression of genes in a subapoptotic manner. Here, we review recent findings on a cooperative antitumor effect when α TOS was combined with immunological agents. It suggests a novel strategy for cancer treatment involving enhanced immune surveillance.
