| - Komplex Leishmania donovani se skládá z L. donovani, L. infantum a L. archibaldi, ačkoli posledně jmenovaný zástupce není považován za validní druh. Rozlišení leishmánií na základě morfologie není možné; používají se biochemické, imunologické a molekulárně biologické přístupy. Multilokální enzymová elektroforéza (MLEE) je v současnosti tzv. zlatým standardem. Sekvenovali jsme geny kodující 5 metabolických enzymů používaných pro MLEE, a to pro rozlišení DNA diverzity příslušných isozymů a rovněž pro posouzení jejich vhodnosti pro multilokální sekvenování na bázi PCR. U 17 kmenů L. infantum, 7 kmenů L. donovani a 3 kmenů L. archibaldi byly sekvenovány geny pro isocitrate dehydrogenázu, jablečný enzym, manosofosfát isolerázu, glukoso-6-fosfát dehydrogenázu a fumarát hydratázu. Fylogenetická analýza založená na 263 kb protein-kodujících sekvencí ukázala silnou korelaci mezi genotypem a geografickým původem. Evropa a Afrika představují na základě našich výsledků nezávislá evoluční centra. (cs)
- The complex Leishmania donovani is comprised of L. donovani (LD), L. infantum (LI) and L. archibaldi (LA). Morphological distinction of species is impractical, so biochemical, immunological and DNA-based criteria were introduced. Multilocus enzyme electrophoresis (MLEE) is the present gold standard. We have sequenced the genes encoding 5 metabolic enzymes used for MLEE, both to resolve the DNA diversity underlying isoenzyme mobility differences and to explore the potential of these targets for higher resolution PCR-based multilocus sequence typing. The genes sequenced were isocitrate dehydrogenase, malic enzyme, mannose phosphate isomerase, glucose-6-phosphate dehydrogenase, and fumarate hydratase, for 17 LI strains, 7 LD strains , and 3 LA strains. Phylogenetic analysis of a concatenated alignment based on a total of 263 kb protein-coding sequences showed strong correlation of genotype with geographical origin. Europe and Africa appear to represent independent evolutionary centres.
