Description
  • Glutamate levels in brain microdialysates of rats with a perinatally induced dysfunction of glutamatergic neurotransmission will be examined to verify its role in etiopathogenesis of schizophrenia-like behaviour of these animals at adulthood. Immunohistochemical and electrophoretic analysis of NMDA receptor subunits in the hippocampi of the above specified rats and schizophrenic brain autopsies will be carried out to further characterize dysfunction of glutamatergic system in this psychosis. Actions of classical (haloperidol) and atypical antipsychotic drugs (clozapine) will be examined in relation to the expression of main NMDA receptor subunits in the rat brain model with a defined dysfunction of glutamatergic. prepulse inhibition of the acoustic startle reaction will be evaluated in relation to the composition of NMDA receptor in the hippocampi of rats with the perinatally induced dysfunction of glutamatergic neurotransmission.   • Hladiny glutamátu v mikrodialyzátech mozku potkanů s perinatálně navozenou dysfunkcí glutamátergního systému budou studovány s cílem ověřit její úlohu v etiopatogeneze této psychóze u těchto zvířat v časné dospělosti. Imunohistochemická a elektroforetická analýza podjednotkové stavby NMDA receptoru v hipokampech výše uvedených potkanů a v autopsiích mozků schizofreniků bude provedena s cílem dále charakterizovat hypofunkci glutamátergního systému u této psychózy. Účinky klasického (haloperidol) a atypického antipsychotika (klozapin) budou vymezeny ve vztahu k proteinové expresi hlavních podjednotek NMDA receptoru v hipokampech potkanů s dysfunkcí glutamátergního systému. Prepulzní inhibice akustické úlekové reakce bude hodnocena ve vztahu k podjednotkové stavbě NMDA receptoru v hipokampu potkana s perinatálně navozenou dysfunkcí glutamátergního neurotransmise.
