Description
| - The mechanism by which some environmental substances called endocrine disruptors (Eds) affect the reproduction of living organisms is still unknown. In this project we will identify the epigenetic changes in reproductive organs after exposure to EDs in vivo and in vitro in the mouse (inbred, outbred) and fish model. Mice and fish will be exposed during embryonic development to EDs at different doses and the transfer of epigenetic defects in the gonads and germ cells in future generations will be monitored. The analysis will focus on changes in DNA methylation and histone posttranslational modification. Analysis of changes in the level of molecules and cells will be carried out on spermatozoa and spermatogonial stem cells. At the level of phenotype changes will be assessed by histopathology of male gonads and sperm analysis. The aim of this project will be to identify and validate biomarkers that mediate inter-generational transfer of male gonadal dysfunction as a result of exposure to selected contaminants in both species. - Stále ještě není známé, jakým mechanismem některé látky životního prostředí tzv. endokrinní disruptory (EDs) ovlivňují reprodukci živých organismů. V tomto projektu budeme na savčím modelu, myši (inbrední, outbrední) a rybím modelu zjišťovat epigenetické změny v reprodukčních orgánech po vystavení EDs v podmínkách in vivo. Myši a ryby budou vystaveny během embryonálního vývoje působení EDs v různých dávkách a bude sledován přenos epigenetických poruch v gonádách a zárodečných buňkách v dalších generacích. Analýza bude zaměřena na změny DNA methylace and histonové posttranslační modifikace. Analýza změn na úrovni molekul a buněk bude prováděna na spermiích, spermatogoniích a zárodečných buňkách. Na úrovni fenotypu bude hodnocena histopatologie samčích gonád a funkční analýza spermií. Cílem tohoto projektu bude zjistit a ověřit biomarkery, které zprostředkovávají mezigenerační přenos samčí gonadální dysfunkce jako důsledek expozice vybraných polutantů u obou druhů živočichů.
