Description
| - The proposed project will deal with spermatozoa ultrastructure, energetic and competition in two groups of model species of chondrostean (Chondrostei) and teleostean (Teleostei) fish. Study on the spermatozoa ultramorphology and ultrastructure in fish, specially focused on structure of axoneme and midpiece, to better understand the roles of mitochondria and basal body structures. This also involves studies of micromechanics and micro hydrodynamics of integrated machinery of axoneme with its associated membrane. Factors reviving motility and fertilizability in %22exhausted%22 spermatozoa of intact and demembranated spermatozoa of fish species. We anticipate the determination of activation/inhibition factors in both fish groups and firstly demonstrate that spermatozoa are able or unable fertilize the eggs with assessment of acrosome integrity. - Předložený projekt se soustřeďuje na řešení ultrasturktury, energie a kompetice spermií u dvou modelových druhů ryb, zahrnující chrupavčité (Chondrostei) a kostnaté (Teleostei) druhy. Studie ultramorfologie a ultrastruktury spermií se zaměří na objasněnípodstaty strukturu bičíku a jeho střední části k pochopení významu struktury mitochondrií a basální části spermií. Znalosti budou využity při studiu mikromechaniky a mikrohydrodynamiky integrovaného mechanismu bičíku a membrány. Dále bude sledován faktorznovuobnovení motility a fertility u tzv. %22vyčerpaných%22 intaktních a demebranovaných spermií. U spermií ryb s obnoveným energetickým potenciál bude zdokumentována schopnnost inhibičních/aktivačních faktorů a opětovného oplození jikry. Mechanismus průnikuspermie bude u chrupavčitých ryb doplněn o studiem akrozomové integrity.
