  • Hard-bodied tick Ixodes ricinus is an important vector of Lyme disease spirochete Borrelia burgdorferi sensu lato. Feeding of ixodid ticks extends over a number of days and it provides an opportunity for vector-host immune interaction. The interplay includes host immune defense against tick feeding as well as modulation of host immune response induced by tick feeding. Recently, increasing evidence supports the idea that tick-borne pathogens take advantage of immunomodulatory factors in tick saliva to facilitate their transmission. We have examined effect of salivary gland extract from I. ricinus on the spirochete proliferation during mouse infection, to test existence of saliva activated transmission (SAT). First direct evidence of B. burgdorferi SAT was demonstrated using real-time PCR on day 1 post infection, when a significant increase in spirochetes was found in mouse tissues, compared with control group.

  • Ixodes ricinus je důležitým vektorem spirochéty Borrelia burgdorferi sensu lato, původce Lymské boreliózy. Klíšťata čeledi Ixodidae sají na hostiteli po dobu několika dnů, což umožňuje vznik vektor-hostitelských interakcí. Toto vzájemné působení zahrnuje imunitní obranu hostitele a zároveň modulaci hostitelské odpovědi vyvolané sáním klíštěte. V současné době stále více imunologických studií podporuje hypotézu, která předpokládá, že patogeny přenášené klíšťaty využívají imunomodulačních molekul ve slinách klíšťat k usnadnění vlastního přenosu do hostitele. Abychom zjistili, zda se slinami aktivovaný přenos (SAT) vyskytuje také u spirochéty B. burgdorferi, sledovali jsme efekt extraktu ze slinných žláz klíštěte I. ricinus na množení borelií v myších tkáních. První den po infekci jsme pomocí real-time PCR zaznamenali signifikantní nárůst spirochét v tkáních myší v porovnání s kontrolou. Tato data představují první přímý důkaz o existenci jevu SAT u B. burgdorferi sensu stricto. (cs)
