Description
| - The project is focused on the synthesis of N6-isopentenyladenine, cis-zeatin, trans-zeatin, N6-benzyladenine, 3-hydroxybenzyladenine and 4-hydroxybenzyladenine derivatives substituted in C2, C8 and N9 positions of the adenine moiety with the appropriate spacer-arm or suitable function group to design potentially biospecific ligands for affinity chromatography of cytokinin derivatives. The proposed project combines our knowledge of organic synthesis of cytokinine derivatives, the ability to study the biological activity of newly prepared derivatives in the extensive number of bioassays and very sensitive and modern quantitative proteomic techniques based on the combination of biochemical and analytical separation methods with highly sensitive mass spectrometry (MALDI-MS, nanoLC-ESI-MSMS) available at the place of work. Results of this project should lead to the identification of new binding and/or storage proteins that can play the crucial role in the regulation of germination and seedling development via modulation of cytokinin levels. - Projekt je zaměřen na syntézu derivátů N6-isopentenyladeninu, cis-zeatinu, trans-zeatinu, N6-benzyladeninu, 3-hydroxybenzyladeninu a 4-hydroxybenzyladeninu, substituovaných vhodným raménkem nebo vhodnou funkční skupinou konkrétně v polohách atomů C2, C8 a N9 adeninového skeletu za účelem designu účinných biospecifických ligandů vhodných pro afinitní chromatografii cytokininových derivátů. Navrhovaný projekt spojuje naše znalosti organické syntézy cytokininových derivátů, schopnost studia biologické aktivity nově připravených derivátů v rozsáhlé řadě biologických testů a zároveň dostupnost velmi citlivých a moderních technik kvantitativní proteomiky, založených na kombinaci biochemických a analytických separačních metod v kombinaci s vysoce citlivou hmotnostní spektrometrií (MALDI-MS, nano-LC-ESI-MSMS) na našem pracovišti. Výsledky z tohoto projektu povedou k identifikaci nových vazebných nebo zásobních proteinů, které mohou hrát klíčovou roli v regulaci klíčení a vývoje semenáčku cestou modulace hladin cytokininů.
