Description
  • DNA replication is fundamental process of living cells that need to overcome the obstacles causing replication to stall or even collapse. Several mechanisms have been proposed to play a role in processing of such lesions, including structure-specific endonucleases. The goal of this proposal is to determine the role of Mus81/Mms4 or its human homolog Mus81/Eme1, during the resolution of stalled and blocked replication forks as well as recombination intermediates. To achieve this goal, we will aim to characterize the effect of Rad54 interaction with Mus81/Mms4 on its nuclease activity. In addition, we will address the role of ATP binding and hydrolysis of Rad54, and determine conservation of this activation in evolution using human homologs. We will also test effect of human RecQ homologs, often associated with genetic disorders (BLM, WRN, RTS syndromes), on human Mus81/Eme1 complex as well as other available nucleases in order to determine their specific role and possible differences in maintaining genomic stability. Finally, we will set up high through put screening for ligands that would specifically inhibit/activate structure-specific nucleases that are required for cleavage of replication and recombination intermediates with the possible aim in therapeutic use.
