  • V souvislosti s možností xenotransplantace prasečích orgánů lidem je zdůrazňováno nebezpečí zoonotické infekce prasečími endogenními retroviry (PERV). Podskupina PERV-A může infikovat lidské buňky pomocí dvou homologních transmembránových proteinů, huPAR-1 a huPAR-2. V této práci jsme charakterizovali lidské PAR a jejich homology u myši a krysy (muPAR a ratPAR). Krysí i myší buňky jsou rezistentní vůči PERV-A. Transfekce huPAR a ratPAR receptorů navodila citlivost k PERV-A u různých resistentních buněk, transfekce muPAR resistenci neovlivnila. Zjistili jsme, že rezistence krysích buněk je daná nízkou endogenní expresí ratPAR. Rezistence myších buněk je způsobena přítomností Pro v pozici 109 v druhé extracelulární smyčce muPAR, který zabraňuje vazbě PERV-A Env na receptor. Z této studie vyplývá, že oba druhy hlodavců jsou rezistentní vůči PERV-A, rezistenci však získaly nezávisle, různými způsoby. (cs)
  • The risk of zoonotic infection by porcine endogenous retroviruses (PERV) has been highlighted in the context of pig-to-human xenotransplantation. A human-tropic PERV subgroup, PERV-A, can enter human cells through either of two homologous transmembrane proteins, huPAR-1 and huPAR-2. Here, we characterised human PARs and their homologues in the PERV-A resistant rodent species, mouse and rat (muPAR and ratPAR). Upon exogenous expression in PERV-A resistant cells, human and rat PARs, but not muPAR, conferred PERV-A sensitivity. Exogenously expressed ratPAR allows PERV-A infection with equivalent efficiency to that of huPAR-1. Endogenous ratPAR expression in rat cell lines appeared to be too low for PERV-A infection. In contrast, in mouse the PERV-A resistance is caused by the presence of Pro at position 109 in the second extracelular loop of muPAR, which affects the PERV-A Env binding to PAR molecules. Both rodent species are PERV-A resistant, however, on different basis.
