  • Photodynamic therapy employs systemic administration of a tumor-localizing photosensitizer followed by local irradiation of the tumor with visible light. The interaction between the excited photosensitizer and endogenous molecular oxygen generates reactive oxygen species, resulting in a cascade of biochemical events leading to cancer cell death. In the present study we investigated photosensitizing properties of two novel mono- and bis-cyclodextrin tetrakis (pentafluorophenyl) porphyrin derivatives in several tumor cell lines and in BALB/c mice bearing transplanted mammary carcinoma. Although both studied porphyrins exhibited similar intracellular localization, the mono-cyclodextrin derivative produced a stronger in vitro phototoxic effect than the bis-cyclodextrin derivative. In vivo, however, the efficacy of the bis- derivative was superior, displaying fast, selective tumor uptake and strong antitumor activity manifested by prolonged elimination of the treated tumors.

  • Fotodynamická terapie představuje systémové podání fotosensitizéru, specificky lokalizovaného v nádoru, následované lokálním osvícením nádoru viditelným světlem. V důsledku interakce mezi molekulami vybuzeného fotosensitizéru a endogenního kyslíku vznikají druhy vysoce reaktivního kyslíku, které spouštějí kaskádu biochemických událostí vedoucích k buněčné smrti. V této studii byly zkoumány fotosensitizující vlastnosti dvou nových mono- a bis-cyklodextrinových derivátů tetrakis(pentafluorophenyl)porfyrinu na několika nádorových buněčných liniích a BALB/c myších s transplantovaným nádorem mléčné žlázy. Oba studované porfyriny byly sice podobně vnitrobuněčně lokalizovány, ale mono-cyklodextrinový derivát měl in vitro silnější fototoxický efekt než bis-cyklodextrinový derivát. In vivo byla naopak fotodynamická účinnost bis-derivátu lepší, což dokazuje jeho rychlé a selektivní vychytávání v nádoru a silná protinádorová aktivita projevující se prodlouženým vymizením nádoru po osvícení.
