  • Hybridní sterilita je běžný postzygotní reprodukční mechanismus, který se objevuje v časných stádiích určení druhu u různých organismů. Mus musculus musculus a Mus musculus domesticus jsou dva nedávno oddělené poddruhy myší vhodné pro genetické studie hybridní sterility. Ukazujeme zde, že vnesení Chr X pocházejícího z M. m. musculus (inbrední kmen PWD/Ph) do genetického základu inbredního kmene C57BL/6J (pocházejícího převážně z M. m. domesticus) způsobuje sterilitu samců. Hybridní sterilita vázaná na chromozom X je spojena se sníženou hmotností varlat, nižším počtem spermií a morfologickými abnormalitami hlaviček spermií. Analýza rekombinantního Chr X u sterilních a fertilních samců spolu s analýzou lokusu kvantitativních znaků (QTL) ukázala, že hybridní sterilita vázaná na chromozom X je podmíněna více geny. Mapování a klonování HstX1 a dalších genů odpovědných za sterilitu samců B6-XPWDYB6 by napomohlo objasnit úlohu Chr X v hybridní sterilitě (cs)
  • Hybrid sterility is a common postzygotic reproductive isolation mechanism, which appears in early stages of speciation of various organisms. Mus musculus musculus and Mus musculus domesticus represent two recently separated mouse subspecies suitable for genetic studies of hybrid sterility. We show that the introgression of Chr X of M. m. musculus origin (PWD/Ph inbred strain) into the genetic background of C57BL/6J inbred strain (predominantly of M. m. domesticus origin) causes male sterility. The X-linked hybrid sterility is associated with reduced testes weight, lower sperm count and morphological abnormalities of sperm heads. The analysis of recombinant ChrXs in sterile and fertile males as well as quantitative trait locus (QTL) analysis of several fertility parameters revealed oligogenic nature of the X-linked hybrid sterility. Mapping and cloning of HstX1 and other genes responsible for sterility of B6-XPWDYB6 males could help to elucidate the role of Chr X in hybrid sterility
