  • Synchronizované kultury zelené řasy Scenedesmus quadricauda byly pěstovány v přítomnosti a absenci 5-fluorodeoxyuridinu, specifického inhibitoru replikace jaderné DNA. Zjišťovali jsme H1 kinázovou aktivitu, akumulaci mitotického cyklínu B a hladinu chloroplastového FtsZ proteinu. Buňky ošetřené FdUrd vykazují vysokou aktivitu mitotické H1 kinázy při absenci jaderného dělení. Kinázová aktivita a akumulace mitotického cyklínu B se plynule zvyšuje do vysokých hodnot bez oscilace. Na rozdíl od jaderného dělení, choroplastové dělení nebylo zablokováno, ale bylo oddáleno. FtsZ struktury byly lokalizovány kolem pyrenoidu a v chloroplastu ve formě mnoha bodů a mini-prstenců. Výskyt a lokalizace FtsZ proteinu byla srovnatelná s kontrolními buňkami, ale u buněk ošetřených inhibitorem se prstence zvětšily, zmnožily a přetrvaly (cs)
  • The synchronized cultures of the green alga Scenedesmus quadricauda were grown in the absence (non-treated cultures) or in the presence (FdUrd-treated cultures) of 5-fluorodeoxyuridine, the specific inhibitor of nuclear DNA replication. A histone H1 kinase activity, mitotic cyclin B accumulation, chloroplast FtsZ protein level were assessed. Interestingly, the FdUrd-treated cells possessed a very high mitotic histone H1 kinase activity in the absence of any nuclear division(s). Compared with the non-treated cultures, the kinase activity as well as mitotic cyclin B accumulation increased continuously to high values without any oscillation. Contrary to nuclear divisions, division of chloroplasts was not blocked but occurred delayed and in a longer time span than in the non-treated culture. FtsZ structures were localized both around pyrenoids and spread inside of chloroplast in form of numerous spots and mini-rings
