Description
| - Selektivní enzymem katalyzovaná hydrolýza oleje ze semen černého rybízu byla studována s cílem získat v jednotlivých třídách lipidů v hydrolyzátu různé koncentrace alfa a/nebo gama-linolenové kyseliny, což by našlo uplatnění ve speciálních dietách. Olej a voda rozpuštěné v superkritickém CO2 tekly náplňovým reaktorem (teplota 40 °C, tlak 15-28 MPa, fiktivní rychlost 0.1-0.7 mm/s) obsahujícím Lipozyme, 1,3-specifickou lipasu z Mucor miehei imobilizovanou na makroporézní pryskyřici. K udržení stabilní aktivity enzymu bylo nutno zabránit srážení vody v reaktoru. Reakce byla řízena jak kinetikou Michaelis-Mentenové, tak i přenosem hmoty. Maximální rychlost uvolňování mastných kyselin na jednotku hmoty enzymu, 2.6 mmol/s kg, byla dosažena při maximálním tlaku a průtoku. Uvolněné kyseliny byly obohaceny alfa-linolenovou, palmitovou a stearovou kyselinou, kdežto v di- a monoacylglycerolech byla zvýšená koncentrace gama-linolenové a stearidonové kyseliny. (cs)
- Selective enzyme-catalysed hydrolysis of blackcurrant seed oil was studied with aim to obtain different levels of alpha- and/or gamma-linolenic acid in lipid classes in the hydrolyzate, making them suitable for special dietary needs. Supercritical CO2 with dissolved oil and water flowed through a packed bed reactor (temperature 40 °C, pressure 15-28 MPa, superficial velocity 0.1-0.7 mm/s) where Lipozyme, a 1,3-specific lipase from Mucor miehei was immobilised on a macroporous ion-exchange resin. The enzyme activity was stable as long as water precipitation in the reactor was prevented. The reaction was controlled by both Michaelis-Menten kinetics and mass transfer. The maximum rate of fatty acids liberation per unit amount of enzyme, 2.6 mmol/s kg, was achieved at the maximum pressure and flow rate. The liberated fatty acids were enriched in alpha-linolenic, palmitic and stearic acids, while di- and monoacylglycerols contained increased levels of gamma-linolenic and stearidonic acids.
