| - To identify amino acid residues responsible for the determination of the yeast Na+/H+ antiporter substrate specificity (ability to recognize K+), the gene encoding the Zygosaccharomyces rouxii Sod2-22 antiporter (non-transporting K+) was mutagenized and a collection of ZrSod2-22 mutants that improved the KCl tolerance of a salt-sensitive S. cerevisiae strain was isolated. Several independent ZrSod2-22 mutated alleles contained the replacement of highly-conserved proline 145 in the fifth transmembrane domain with a residue containing a hydroxyl group (serine, threonine). The ZrSod2-22p with P145S or P145T substitution mediated, in contrast to the wild-type antiporter, transport of K+. Simultaneously, the P145S(T) mutation decreased the antiporter transport capacity for both Na+ and Li+. Thus, the Pro145 was found to be a critical residue for the yeast plasma membrane antiporters’ function, as far as the substrate specificity range and transport activity are concerned

- Aby bylo možné identifikovat aminokyselinové zbytky odpovědné za určení substrátové specifity (rozpoznání K+) u Na+/H+ antiporterů kvasinek, byla provedena mutageneze genu kódujícího antiporter Sod2-22 z kvasinky Zygosaccharomyces rouxii (netransportující K+) a byla izolována kolekce mutantních verzí ZrSod2-22p, jejichž exprese v Saccharomyces cerevisiae (v mutantním kmeni citlivém k solím) vedla ke zvýšené toleranci buněk ke KCl. U několika nezávisle získaných mutantních alel ZrSod2-22 došlo k záměně konzervovaného zbytku prolinu 145 v páté transmembránové doméně za aminokyselinový zbytek obsahující hydroxylovou skupinu (serin, threonin). ZrSod2-22 antiporter obsahující mutaci P145S nebo P145T byl schopen, na rozdíl od nemutovaného přenašeče, transportovat K+. Současně mutace P145S(T) snižovala transportní kapacitu antiporteru pro kationty Na+ a Li+.
