  • We constructed a reporter gene system, based on luciferase genes from Vibrio harvei, for measurement of translation nonsense suppression in Streptomyces. Streptomyces lividans TK 24 was transformed with the plasmids and the reporter system was tested by growth of the strain in the presence of streptomycin as a translation accuracy modulator. Streptomycin increased nonsense suppression on UAA nearly 10-fold and more than 20-fold on UAG. Whereas on UGA, the most frequent stop signal in Streptomyces, the effect was negligible

  • Sestrojili jsme genový reporterový systém využívající luciferázu z Vibrio harvei pro měření translační nonsense suprese u streptomycet. Transformovali jsme Streptomyces lividans TK 24 plasmidem nesoucím reporterový systém, který jsme testovali během růstu kultury v přítomnosti a nepřítomnosti streptomycinu jako modulátoru přesnosti translace. Streptomycin zvyšoval nonsense supresi UAA téměř desetkrát a v případě UAG více než dvacetkrát. V případě UGA, nejčastějšího stop signálu u streptomycet, byl efekt zanedbatelný (cs)
