  • Rpg1p/Tif32p/eIF3a je velká podjednotka eIF3 komplexu fungujícího v iniciaci translace v Saccharomyces cerevisiae.Zjistili jsme, že v nestresovaných buňkách obě, jak GFP tak i mRFP fůze Rpg1p/eIF3a byly rovnoměrně distribuovány v cytoplazmě. Po teplotním šoku Rpg1p/eIF3a akumuloval ve fluoreskujících granulích ve vrcholu pupene a v krčku. Tvorba granulí nebyla závislá na přítomnosti mikrotubulů a aktinu, a nezávisí na Sla2. Předpokládáme, že Rpg1p/eIF3a zprostředkuje tvorbu a rozpuštění stresových granulí. (cs)
  • Rpg1p/Tif32p/eIF3a is the large subunit of the eIF3 core complex involved in translation initiation in Saccharomyces cerevisiae.We found that in unstressed cells Rpg1-GFP fusion is uniformly distributed throughout the cytoplasm. Upon osmotic upshift the protein rapidly accumulated in fluorescing foci dispersed in the cytoplasm. Accumulation of Rpg1-GFP fusion was observed near the tip of small buds and at the mother-daughter neck. The formation of Rpg1-GFP granules was not altered by disruption of microtubules or F-actin, and by the absence of Sla2p.We suggest that Rpg1p/eIF3a mediates formation and dissolution of stress granules.
