  • Two methods for monolayer immobilisation of photosystem II isolated from the thermophilic cyanobacteria Synechococcus elongatus and prospects for its use as a biosensor for detection of herbicides are reported: (i) Immobilisation based on recombinant (His)6-tagged PSII coupled with Ni-NTA chelator monolayer on Au electrode. (ii) Immobilisation based on the natural PSII coupled with the protein A - anti-D1 antibody modified Au electrode. Here, CYS-SAM-NTA-PSII monolayers were compared with traditional BSA-GA-PSII cross-linked gel matrix and better performances of the derived electrochemical biosensors were pointed out. Better diffusion of inhibitors and mediators resulted in improved sensitivity, velocity of the response and lower I50 for herbicides

  • Práce se týká dvou metod imobilizace monovrstev fotosystému II z termofilní sinice Synechococcus elongatus a výhled jejich využití v biosenzorech pro detekci herbicidů: (i) Imobilizace založená na rekombinantním fotosystému II s šestinásobnou histidinovou kotvou spojenou přes NI-NTA chelator ke zlaté elektrodě. (ii) Imobilizace založená na přírodním fotosystému II navázaném zlatou elektrodu modifikovanou spojením proteinu A s D1protilátkou. V této práci jsou monovrstvy CYS-SAM-NTA-PSII srovnávány s tradiční imobilizací v zesíťované matrix BSA-GA-PSII a jsou zdůrazněny lepší provedení modifikovaného elektrochemického senzoru.Lepší difúze inhibitorů a mediátoru mají za následek zlepšenou citlivost a rychlost odezvy a nižší I50 pro herbicidy (cs)
