  • Complex of OsO4 with 2,2’-bipyridine was used as a probe of DNA structure and an electroactive marker of DNA in hybridization sensors. It produces several voltammetric signals, most negative of them only at Hg electrodes. This signal is of catalytic nature due to evolution of H2 in voltammetry of DNA modified by complex of OsO4 with 2,2’-bipyridine (DNA-Os,bipy). Solid amalgam electrodes modified with Hg menisci of Ag (m-AgSAE), Cu (m-CuSAE), Au and combined Bi and Ag, were used. Besides hanging Hg drop electrode (HMDE), catalytic current was observed only on m-AgSAE and m-CuSAE, not on Au and Bi amalgams. Detection limit of DNA-Os,bipy on HMDE was 0.1 ng mL-1 (RSD = 2.3%, N = 11), and on m-AgSAE 0.2 ng mL-1 (RSD = 3.1%, N = 11). m-AgSAE was successful in double-surface DNA hybridization, offering highly specific discrimination between complementary (target) and nonspecific DNAs, as well as for determination of length of a repetitive DNA sequence.

  • Komplex 0s04 s 2,2´-bipyridinem byl použit jako sonda struktury DNK a jako elektroaktivní značka DNK v hybridizačních čidlech. Ve voltametrii dává několik signálů, nejnegativnější z nich jen na Hg elektrodách. Ten je katalytické povahy, způsobený vylučováním H2 při voltametrii DNK modifikované komplexem OsO4 s 2,2´- pyridinem (DNK-Os,bipy). Byly použity pevné amalgámové elektrody, modifikované rtuťovým meniskem, z Ag (m-AgSAE), z Cu (m-CuSAE), z Au, a kombinované z Bi a Ag. Vedle visící rtuťové kapkové elektrody (HMDE), katalytické proudy byly pozorovány jen na m-AgSAE a m-CuSAE, nikoli na amalgámech Au a Bi. Detekční limit DNA-Os,bipy na HMDE byl 0.1 ng mL-1 (RSO = 2,3%, N = 11) a na m-AgSAE 0,2 ng mL-1 (RSO = 3,1%, N = 11). m-AgSAE byla úspěšná při zjišťování dvoupovrchové DNK hybridizace, umožňující vysoce specifické rozlišení mezi komplementární (cílovou) a nespecifickou DNK, a stanovení délky opakované sekvence DNK.
