Description
| - Although the adducts of various chemical carcinogens with DNA have been studied extensively, little is known about their excretion in the urine, which can be used for a non-invasive assessment of the overall DNA adduct formation in the body. For example, no DNA adducts arising from exposure to benzene, a ubiquitous industrial and environmental genotoxic carcinogen, have been found in the urine as yet. Here we wished to detect in mouse urine the DNA adducts derived from the benzene metabolite p-benzoquinone (BQ), which was reported to react with 2´-deoxyadenosine, 2´-deoxyguanosine and 2´-deoxycytidine in double stranded DNA (but not in tumour cells or in vivo) to give cyclic adducts, identified as 9-hydroxy-1,N6-benzeno-2?-deoxyadenosine (DABQ), 7-hydroxy-1,N2-benzeno-2?-deoxyguanosine (DGBQ) and 3-hydroxy-3,N4-benzeno-2?-deoxycytidine (DCBQ), respectively.
