  • The study of the role of polyphenols in malt, hops and beers is very important and therefore new rapid extraction techniques are required. Modern Pressurized Fluid Extraction (PSE) technique coupled with HPLC and high sensitivity CoulArray detection was used for analysing of some of the polyphenols in malt and hops. The qualitative and quantitative dependence of extracted polyphenols on extraction temperature was found under optimized extraction conditions of PSE technique. Recoveries within the temperature interval 40–60 °C were constant with the exception of ferulic acid. Outside of this interval concentrations of some polyphenols depend on the temperature significantly. Obtained results were compared with results obtained by the currently used warm water extraction methods. This PSE method together with HPLC and high sensitivity CoulArray detector could be a basis for standard analytical methods which could be independent on any enzymatic activities.

  • Studium úlohy polyfenolů ve sladu, chmelu a pivu nabývá na významu, a proto je kladen důraz na vývoj nových extrakčních technik. Moderní tlaková fluidní extrakce (PSE) ve spojení s HPLC a vysoce citlivým CoulArray detektorem byla užita pro analýzu polyfenolů ve sladu a chmelu. Za optimalizovaných PSE podmínek byla sledována extrakční závislost na teplotě. Výtěžnosti v intervalu 40-60 oC byly konstantní s výjimkou kyseliny ferulové. Mimo tento interval získané koncentrace některých polyfenolů byly teplotně značně závislé. Získané výsledky byly porovnány s výsledky změřenými při dosud používaných extrakcích do horké vody za varu (chmel) nebo při rmutování (slad). Extrakční PSE metoda společně s HPLC a vysoce citlivým CoulArray detektorem by mohla být základem pro standardní analytické metody, které by byly nezávislé na enzymatických aktivitách. (cs)
