  • Metabolic engineering of the flavonoid pathway may be used for modifying nutritional, pharmaceutical and ornamental properties of plants. The PAP1 (AtMYB75) is a R2R3 MYB family member and stimulates the expression of flavonoid pathway genes in Arabidopsis thaliana . Previously, PAP1 was introduced into Humulus lupulus plants by Agrobacterium-mediated genetic transformation. Employing quantitative PCR, we show that each transgenic plant harbors one copy of PAP1. The expression of the structural genes CHS_H1, CHI, and F3'H, essential for the biosynthesis of flavonoids, was elevated in PAP1 transgenic plants compared to the wildtype hop. Chemometric methods were successfully used to discriminate between wildtype and transgenic plants expressing the transcription factor PAP1. Thus, transcription factor PAP1 activated the expression of these three genes in transgenic plants, opening the possibility for improving the content of pharmaceutically important secondary metabolites in hop.
