Description
  • Určili a analyzovali jsme konzervativní oblasti minikroužků z různých rostlinných trypanosomatid. Cílem této studie bylo najít novou metod pro rozlišování různých trypanosomatid vyskytujících se u rostlin, konkrétně bičíkovců nacházejících se v míze, jiných druhů vyskytujících se v latiferozních pletivech a konečně druhů, jejichž výskyt je omezen na plody, semena a květy. Posledně jmenované druhy se nevyskytují v samotných rostlinách a jsou považovány za oportunní parazity. Jelikož jsou někteří trypanosomatidi parazitující floem rostlin původci závažných chorob, je velmi důležité určit přesné definice typů těchto bičíkovců. Konzervativní oblast minikroužků poskytuje řadu specifických rysů v poměrně malé oblasti, a to zejména tři funkční elementy nezbytné pro replikaci minikroužků. Fylogenetické stromy vytvořené na bázi minikroužkových analýz se značně podobají stromům založených na izoenzymech, RAPD a jednotlivých genových sekvencích, čímž je potvrzena validita naší analýzy.   • The sequences of minicircle conserved regions from various plant trypanosomatids have been determined and analyzed. The goal of this study was to add another tool to the arsenal of molecular probes for distinguishing between the different trypanosomatids occuring in plants: systemic trypanosomatids multiplying in the sap, those from the laticiferous tubes, and those developing in fruits, seeds or flowers but not in the plant itself and that are frequently considered as opportunistic insect trypanosomatids. As some plant intraphloemic trypanosomatids are the causative agents of important diseases, a clear definition of the different types of trypanosomatids is critical. The conserved region of the mitochondrial minicircle provides several sequence-specific features. Trees generated from the analysis recapitulated trees drawn from analyses of isoenzymes, RAPD, and particular gene sequences, supporting the validity of the small region used in this work.
