  • The effect of several metal cations Cu2+, Cd2+, Co2+, Ba2+) was tested in attempts to improve crystallization procedure and verify a newly discovered cross-crystallization method with two selected proteins; di-heme cytochrome c4 from anaerobic purple sulphur bacterium Thiocapsa roseopersicina and sweet-tasting protein thaumatin from the African berry Thaumatococcus daniellii. Presence of Cu2+ ions promoted dramatic improvement in crystal morphology, internal packing and diffraction quality. This investigation qualitatively established the influence of cupric cations on the crystal growth by using the cross-crystallization procedure.   • Efekt vybraných kationtů kovů (Cu2+, Cd2+, Co2+, Ba2+) byl testován ve snaze zlepšit krystalizaci a ověřit nově vyvinutou krystalizační metodu nazvanou %22cross-crystallization alias cross-influence procedure (CIP)%22. Testovány byly dva proteiny; cytochrom c4 z anaerobní sulfurické bakterie Thiocapsa roseopersicina a sladký protein thaumatin z Thaumatococcus daniellii. Divalentní Cu2+ způsobily nejdramatičtější zlepšení morfologie, vnitřního uspořádání krystalů a difrakční kvality. Toto zjištění pomohlo kvalitativně popsat vliv kationtů mědi na růst krystalů použitím CIP. CIP je založena na použití sady aditiv přidávaných do kapek, které vykazují pozitivní vliv na rezervoár tím, že citlivě mění pH a tlak vypařování vody nad sousedící kapkou obsahující protein. V případě thaumatinu bylo zjištěno, že vliv Cu2+ iontů podněcuje evidentně odlišnou vnější morfologii a vnitřní uspořádání. V případě cytochromu c4, krystaly v difrakční kvalitě byly získány pouze při použití CIP. Krystaly byly vhodné pro (cs)
