Description
  • Applicability and conditions for in vitro cultures for testing of pome fruit (apple, pear) resistance to fire blight were investigated. Verified isolate of Erwinia amylovora (Ea 8/95) isolated from necrotic tissues of hawthorn was used in tests. Three methods of artificial inoculation of in vitro shoot cultures were tested: 1) removal of apical part of the shoot tip with scissors and applying 3 µl of a pathogen suspension with a pipette on the cut surface, 2) crushing the leaf blade with forceps dipped in a bacterial suspension, and 3) injecting 250 µl of bacterial suspension into the cultivation vessel containing in vitro shoot tip cultures. Out of the tested cytokinins, only media with phytohormone BAP proved to be useful for artificial inoculation with E. amylovora. Abundant callus formation and swollen shoots were observed in media with TDZ. These callused shoots could not be used directly in artificial inoculation studies. Medium with phytohormone 2iP did not promote satisfactory proliferation in our experiments. Removal of the shoot tip with scissors and applying inoculum on the cut area (method 1) proved to be the best method for artificial inoculation. The remaining two methods were not suitable. Concerning the method 2, an injury of plants by forceps occurred. In the case of the method 3, excessive contamination of tested material and cultivation media was noted. Preliminary results of inoculation of in vitro cultures by E. amylovora indicate the possibility of using of the method for testing of pome fruit resistance against fire blight. The advantage of in vitro culture testing is the possibility to test all year round in an area of only several square meters. Moreover, the influence of rootstock and interstem on the level of resistance of tested cultivar is eliminated.
