  • In this research was developed in-house GMO chip for identification of four plants species, five genetically modified organisms (GMO) and three GMO screening elements. The main goal was to get signal after hybridization of labeled total genomic DNA on the chip. It was tested three ways of labeling templates using fluorescent Cy3/Alexa3 and Cy5/Alexa 5 colours, it was tested hybridization of PCR products, total genomic DNA and treated DNA on the chip. It was determinated sensitivity of developed chip in a way of dilution of hybridization teplate. The value of 25 ng DNA was find as a required amout of DNA, who give a significant signal. However, the signal after hybridization of total genomic DNA wasn’t significant. Thus, we propose another procedures for signal improvement of total genomic DNA.

  • Tento příspěvek popisuje GMO chip pro identifikaci čtyřech rostlinných druhů, s pěti genetickými modifikacemi a tři GMO screeningové elementy. byly testovány tři postupy značení celogenomové DNA.byly použity fluorochromy Cy3/Alexa3 a Cy5/Alexa 5, hybridizace PCR prosuktů. Signálu bylo dosaženo při použití 25ug DNA. Specificita signálu však nebyla dostačující, proto jsme navrhli jiný způsob značení DNA. (cs)
