Description	The aim of the proposed project is development of a stable phage preparation containig as a effective compound the novel phage mutants with lytic activity towards multidrug resistant Staphylococcus aureus strains. The lysis-optimized host-range mutants of the virulent phage 812 will be prepared by genetic selection. The lytic activity of the new mutants will be characterized in detail on a representative set of recent clinical S. aureus strains. Genomic analysis will be used to exclude presence of toxic genes in genomes of therapeutic phages. Genetic basis of host-range mutations will be determined by analysis of restriction-modification systems and prophage genes influencing sensitivity of host S. aureus. The study will be complemented by analysis of phage- and bacterial protein composition of phage lysates to assess imunogenicity of phage preparations. (en) Cílem projektu je příprava stabilního terapeutického preparátu, jehož účinnými složkami budou nové fágové mutanty s lytickým účinkem na patogenní kmeny Staphylococcus aureus multirezistentní k antibiotikům. Mutanty polyvalentního bakteriofága 812 s širokým rozmezím hostitelů budou připraveny prostřednictvím genetické selekce. Lytická aktivita nových mutant bude detailně charakterizována na reprezentativním souboru klinických izolátů S. aureus. Pro vyloučení přítomnosti genů virulence v genomech terapeutických bakteriofágů bude použita genomická analýza. Genetický základ mutací v rozmezí hostitele bude stanoven analýzou restrikčně modifikačních systémů a profágových genů ovlivňujících citlivost hostitelských kmenů S. aureus. Výzkum bude doplněn analýzou fágových a bakteriálních proteinových složek fágových lyzátů pro posouzení imunogenních vlastností fágového přípravku.
