Description
| - Within this project, new approaches for the detection and typing of the causative agent of paratuberculosis using culture and molecular biology methods base on the PCR will be introduced. The project will trace sources and distribution of M. a. paratuberculosis and analyse the risks and transmission ways among domestic and wild living ruminants. In selected herds, infection intensity of excrements (solid and liquid manure etc.) from animals infected with the causative agent of paratuberculosis, its survival and risk of contamination of fertilized soil and pastures will be investigated. In the experiments, potential penetration of the causative agent of paratuberculosis via root system into the tissues of vegetables cultivated in infected soil will be assessed. Within evaluation of milk and milk product, contamination risks during their processing, presence of live, sublethally damaged or dead cells of the causative agent of paratuberculosis in biofilms prepared in vitro and biofilms from selected dair (en)
- Prověřit zdroje M. a. paratuberculosis a prokázat výskyt a přežívání v půdě, hnojivech, rostlinách, v mléce a mléčných výrobcích a v biofilmech mléčných provozů. Pro tyto účely použít stávající metody kultivace, PCR a RFLP a vyvinout nové postupy.

| - The project aim was to verify the M. paratuberculosis sources and to demonstrate its presence and survival in soil, fertilizers, plants, milk and dairy products as well as in the biofilm present in dairy operations. The existing cultivation methods and the PCR and RFLP methods were used to attain this project objective and new procedures were developed. The outputs anticipated were fulfilled. (en)
