Description	The subject of this project is analysis of the PALB2 gene role in a pathogenesis of breast, ovarian and pancreatic carcinoma in the Czech Republic. We will perform mutation analysis in a group samples from breast cancer patients from high-risk families BRCA1/2 negatively tested, in samples of additional risk groups of breast cancer, sporadic breast or ovarian and samples of unselected pancreatic cancer. Mutation analyses will be performed by standard techniques (HRM, dHPLC, sequencing, MLPA) well-established in our laboratory. A frequency of pathogenic mutations in patients will be compared with a frequency in control samples and statistically evaluated. Inactivation of wild-type allele in a tumor tissue using LOH analysis and hypermethylation of promoter region will be studied in samples with identified PALB2 alterations. Impact of unclassified variants of the PALB2 gene on its protein product function will be studied using in vitro functional assays in stable breast cancer cell lines. (en) Předmětem předkládaného projektu je analýza úlohy genu PALB2 v patogenezi karcinomu prsu, ovaria a pankreatu v České Republice. Bude provedena mutační analýza tohoto genu u vzorků od pacientek s karcinomem prsu z vysoce rizikových rodin bez mutace v genech BRCA1/2, u vzorků od dalších rizikových skupin pacientů s karcinomem prsu a ovaria, u vzorků sporadického karcinomu prsu nebo ovaria a u vzorků neselektovaného karcinomu pankreatu. Mutační analýzy budou provedeny standardními technikami (HRM, dHPLC, sekvenování, MLPA) rutinně zavedenými v naší laboratoři. Frekvence patogenních mutací bude srovnána s frekvencí v kontrolní populaci a statisticky vyhodnocena. U pacientů s nalezenými alteracemi genu PALB2 bude studována inaktivace druhé alely v nádorové tkáni pomocí analýzy LOH a hypermetylace promotorové oblasti. Pomocí in vitro analýz na modelech stabilních standardně kultivovaných buněčných linií ca prsu (MCF-7) bude zkoumán vliv alterací genu PALB2 na funkci jeho proteinového produktu.
