The study will be accompanied by the analysis of Hsp90 and its isoforms alpha and beta, their posttranslational modifications, CHIP and HOP expression in breast, colon and ovarian cancers and determination of the role of Hsp90 in stabilization of target proteins. This analysis could predict the potential therapy response to Hsp90 inhibitors. The analysis of most efficient type of chemotherapy in tumor cell lines with hyper-activated Hsp90 will identify the constituents of cancer-relevant chaperone pathways and unravel their mode of action. The p53 expression level, its functional status and chaperone status in tumor cells will be analyzed to understand whether over-expression of mutant p53 together with Hsp90 hyper-activation is related to response to chemotherapy. Bude provedena analýza exprese Hsp90 a jeho izoforem alfa a beta, jejich posttranslačních modifikací a exprese proteinů CHIP a HOP v nádorech prsu, střeva a vaječníku a studována úloha Hsp90 při stabilizaci cílových proteinů. Získané výsledky tak umožní předpovídat odpověď na potenciální terapii založenou na inhibitorech Hsp90. Nalezení nejefektivnějšího typu chemoterapie na modelu nádorových buněk s hyperaktivovaným Hsp90 pomůže identifikovat jednotlivé součásti signálních drah, které určují mechanismus působení chaperonů v nádorech. Bude studována úroveň exprese p53, zároveň s jeho funkčností a stavem chaperonového systému v nádorových buňkách s cílem pochopit, nakolik zvýšená exprese mutantních p53 spolu se současnou hyperaktivací Hsp90 souvisí s odpovědí na chemoterapii.
