Description	It is proposed to apply modern electrochemical techniques in connection with mercury abd solid electrodes in studies of covalent and/or non-covalent interactions of nucleic acids (NA) and proteins (including tumour supressor protein p53) with selected carcinogens, mutagens or anti-cancer drugs. 1. Electroactive covalent adducts of biomacromolecules with these substances will be electrochemically characterized. 2. Interactions of biopolymers with the above mentioned substances will be studied using biopolymer-modified electrodes. 3. Specific interactions of unmodified and modified nucleic acids with proteins (including DNA covalent adduct-specific antibodies, enzymes and protein p53) will be studied. Results of the electrochemical studies will be compared with other methods, including optical, chromatographic, DNA sequencing and immunochemical techniques. (en) Navrhujeme využít moderní elektrochemické metody ve spojení s rtuťovými a pevnými elektrodami při studiu kovalentních a nekovalentních interakcí nukleových kyselin a bílkovin (včetně nádorového supresoru p53) s vybranými karcinogeny , mutageny a protinádorovými léčivy. 1. Elektroaktivní kovalentní adukty biomakromolekul s těmito látkami budou charakterizovány elektrochemicky. 2. Interakce biopolymerů s uvedenými látkami budou studovány s využitím elektrod modifikovaných biopolymery. 3. Budou studovány specifické interakce modifikovaných a nemodifikovaných nukleových kyselin s proteiny (včetně protilátek specifických pro kovalentní adukty, enzymů a proteinu p53). Výsledky elektrochemických měření budou porovnány s jinými metodami, včetně optických, chromatografických, sekvenačních a imunochemických technik.
