Description
| - Both circadian and endocrine mechanisms have been implicated in aging of model organisms, but their interrelations are not known. Previous studies on the linden bug, Pyrrhocoris apterus, defined roles of endocrine tissues, neurosecretory cells of the brain, producing insulin-like proteins, and the corpus allatum (producing juvenile hormone, JH), in the regulation of aging and showed that changes in adult longevity caused by JH are associated with changes in the expression levels of two circadian clock genes in the fat body. These results suggest a link between the circadian clocks and the endocrine regulation of longevity. To test this hypothesis, life span will be manipulated by environmental and endocrine interventions and age related changes in the expression of several clock genes will be monitored. To discriminate between common and tissue specific responses of clock genes to hormonal changes, clock gene expression will be compared between two peripheral tissues, the fat body and the gut. These tissues show different physiological responses to JH.. Endocrine interventions will be performed by means of microsurgery. Gene expression will be determined by real time PCR and in situ hybridization, RNA interference will be used for gene silencing. (en)
- Jak cirkadiánní, tak endokrinní mechanizmy se účastní řízení stárnutí modelových organizmů, ale jejich vzájemný vztah není znám. Předchozí studie na ploštici Pyrrhocoris apterus definovaly úlohu endokrinních tkání, neurosekrečních buněk mozku, produkujících insulinu podobné proteiny, a corpus allatum, produkujícím juvenilní hormon (JH) v řízení stárnutí, a ukázaly, že změny v délce života dospělců způsobené JH jsou spojeny se změnami v expresi dvou cirkadiánních genů v tukovém tělese. Tyto výsledky naznačují vazbu mezi cirkadiánními hodinami a endokrinním řízením stárnutí. Abychom ověřili tuto hypotézu, budeme manipulovat délku života environmentálními a endokrinními intervencemi a budeme monitorovat změny v expresi několika cirkadiánních genů během stárnutí. Abychom odlišili obecné a tkáňově specifické odpovědi cirkadiánních genů na hormonální změny, exprese cirkadiánních genů bude porovnána mezi dvěma periferními tkáněmi, tukovým tělesem a střevem. Tyto tkáně se liší ve fyziologické odpovědi na JH. Endokrinní intervence budou prováděny pomocí mikrochirurgie. Exprese genů bude analyzována pomocí RT PCR a in situ hybridizace, RNA interference bude použita pro potlačení exprese genů.
