Description	Oligosaccharide-mediated recognition and adhesion are key points in the early steps of infection of many pathogens. The project is focused mainly to structure-function studies of carbohydrate-binding proteins from Pseudomonas aeruginosa and Burkholderia cenocepacia. Infections with both pathogens often represent a life-threatening event in immunocompromised patients. Chronic lung colonization by P. aeruginosa is the major cause of cystic fibrosis patients mortality, 20% of infection by B. cenocepacia end in fatal necrotising pneumonia.Selected proteins will be cloned and expressed in recombinant form. The fine specificities will be studied by binding experiments using ELLA, microcalorimetry and SPR methods with oligosaccharides of biological interest, such as blood group antigenic determinants, mucin fragments and alginate oligosaccharides. Microcalorimetry with the best ligands will be performed in order to determine the thermodynamics of binding. Crystal structure determination of (en) Specifické rozpoznávání a adheze založené na interakcích s oligosacharidy jsou často prvotním stadiem infekce mnoha patogenů. Projekt je zaměřen především na strukturně-funkční studie proteinů vážících sacharidy z Pseudomonas aeruginosa and Burkholderia cenocepacia. U pacientů s oslabenou imunitou představuje infekce těmito patogeny často život ohrožující proces. Chronická kolonizace plic P. aeruginosa je hlavní příčinou úmrtí u pacientů s cystickou fibrosou, 20 % infekcí B. cenocepacia končí fatální nekrotickou pneumonií.Vybrané proteiny budou klonovány a exprimovány v rekombinantní podobě. Specificity vazebných interakcí budou studovány pomocí metod ELLA a SPR s biologicky důležitými oligosacharidy, jako jsou antigenní determinanty krevních skupin,fragmenty mucinů a oligosacharidy alginátů. Pro zjištění termodynamiky vazby budou s nejlepšími ligandy prováděna mikrokalorimetrická měření.
