Description
| - The project is focused on synthesis and study of new ligands and their complexes with lanthanides and transition metals with aim to participate on the international research and development of new contrast agents for Magnetic Resonance Imaging Synthesis of new ligands derived from tetraazacycles bearing acetate, methylphosphonate or phosphinate, eventually, other pendants would be the main part of the project. Through the pendant groups, additional functionalities able to anchor biomolecules or to change relaxivity in dependence on pH, concentration of calcium etc., will be bond to the ligands. In addition to preparation of the model complexes and determination of their structure by x-ray diffraction methods, research will be directed to the solution studies. Thermodynamic (stability constants, isomerism) as well as kinetic (formation and dissociation rates of complexes, water exchange rate etc.) properties of the complexes will be investigated on the parent compounds. - Projekt je zaměřen na syntézu a studium nových ligandů a jejich komplexů s lanthanoidy a s přechodnými kovy s cílem se podílet na mezinárodním výzkumu a vývoji nových kontrastních látek pro tomografii NMR. Hlavní součástí by měla býti syntéza nových ligandů odvozených od tetraazacyklů nesoucích acetátové, methylfosfonátové nebo fosfinátové, případně další pendantní skupiny. Prostřednictvím pendantních skupin budou k ligandům připojeny další funkční skupiny schopné se vázat na nosič a také na biomolekuly případně měnit relaxivitu gadolinitých komplexů se změnou pH, koncentrací vápníku atp. Vedle přípravy modelových sloučenin a určení jejich struktury metodami RTG difrakce, bude pozornost soustředěna na roztokové studie. Termodynamické (konstanty stability, isomerizace) i kinetické (rychlosti tvorby a disociace komplexů, rychlost výměny vody atd.) vlastnosti budou zkoumány na vybraných komplexech jednodušších ligandů.
