| Description
| - The aim of this project is the development of the methodology of synthesis of purine derivatives bearing diverse highly functionalized C-substituents in the position 6, application of the methodology for the synthesis of novel types of substituted purinebases and nucleosides, as well as their preliminary biological (cytostatic, antiviral) activity screening. Several approaches towards the synthesis of the title compounds will be studied: i) cross-coupling reactions of halopurines with protected ormasked functionalized organometallics, ii) cross-coupling of halopurines with enolates derived from ketones, carboxylates and amino acids, iii) generation of (purin-6-yl)magnesium or -zinc reagents and their reactions with electrophiles and iv)[2+2+2]-co-cyclotrimeriztions of 6-ethynylpurines with diynes. - Cílem projektu je vývoj nových metod syntézy purinových derivátů nesoucích různé funkcionalizované C-substituenty v poloze 6, aplikace této metodiky pro syntézu nových typů substituovaných purinových bází a nukleosidů a předběžné studium jejichbiologické (cytostatické, protivirové) aktivity. Pro syntézu cílových látek budou studovány následující postupy: i) cross-coupling reakce halopurinů s chráněnými nebo maskovanými funkcionalizovanými organokovy, ii) cross-coupling reakce halopurinůs enoláty odvozenými od ketonů, karboxylátů a aminokyselin, iii) generování (purin-6-yl)hořečnatých nebo zinečnatých činidel a jejich reakce s elektrofily a iv) [2+2+2]-ko-cyklotrimerizace 6-ethynylpurinů s diyny. Každý z těchto postupů by měl být vhodnýpro přípravu různých typů substituovaných purinů. Kromě syntézy a studia biologické aktivity nových purinových derivátů be tento projekt měl přispět k obecnému vývoji metodiky cross-coupling reakcí a purinové chemii.
